G protein-coupled receptors (GPCRs) are among the most important drug targets, and sales from drugs targeting GPCRs exceed sales from drugs targeting other protein classes (details). Most of the GPCRs used for drug screening and structural studies are produced using either mammalian or baculovirus protein expression systems. GlaxoSmithKline scientists expressed 16 GPCRs using a baculovirus system and concluded that overall performance was superior to other systems (Akermoun M. et al., Protein Expr. Purif., 44: 65-74, 2005). AB Vector offers the largest selection of recombinant baculoviruses for producing the G protein-coupled receptors of top priority. We routinely provide 1ml stocks, but larger amounts could be made available contingent on the demand.

GPCRs have proven to be eminently suitable for therapeutic intervention, in part due to the fact that they are easily accessible to the drugs on the cell surface. Among approximately 500 drugs on the market about 50% target GPCRs. Unfortunately, GPCRs are among the most difficult proteins to produce. Due to difficulties in producing native GPCRs in sufficient quantities, not a single structure of a therapeutically relevant GPCR was solved, thus impeding protein-structure based drug design for these targets.

AB vector believes that GPCR expression levels are low due to insufficient quantities of protein folding factors specific to GPCRs. For example, a mammalian or baculovirus system is universally expected to produce any human protein. However, many proteins misfold and are either degraded or produced in the form of biologically inactive aggregates. The root of the problem is that expression systems are geared for producing target proteins in much higher than physiological amounts without providing sufficient quantities of corresponding protein folding partners.

AB Vector succeeded in a many-fold improvement in the yield of steroid receptors by providing an ensemble of protein folding factors specific to these receptors (Multi-chaperone systems). We hope to achieve similar improvement in the production of GPCRs by providing ensembles of protein folding factors specific to GPCRs.
